Optimised isolation, characterisation and osteogenic induction of mesenchymal stem cells from umbilical cord blood: comparison with bone marrow mesenchymal and embryonic stem cells

Applications are invited for the above post to work on an EU Framework VI-funded working with Dr Lee Buttery in the Division of Drug Delivery and Tissue Engineering. This project is part of a large multi-centre, multidisciplinary project to optimise the isolation and expansion of mesenchymal stem cells from human umbilical cord blood. The pluripotency of these cells will be examined, but lineage-specific differentiation will be focused on osteogenesis. The cells will be characterised by genomic, proteomic and bioimpedance profiling, pre- and post-osteogenic differentiation. A key part of the project will be comparison of cord blood mesenchymal stem cells with mesenchymal isolated from human bone marrow and also embryonic stem cells. The work at Nottingham will focus on the osteogenic differentiation of human ES cells.

Full bioinformatics integration of datasets will allow us to identify specific and/or novel signalling factors associated with these stem cells. Comparative analyses of the population doubling times, telomere length and telomerase activity will identify the longevity of the stem cells. Novel expansion techniques will be combined with scale-up procedures and the generation of cord blood mesenchymal stem cell lines for banking using optimised cryopreservation protocols. In vitro and in vivo biocompatibility assays using a range of biomimetic scaffolds will be exploited, complementing in vivo homing, engraftment and bone repair models.

Our integrated approach using existing and complementary expertise will provide a timely and thorough evaluation of cord blood mesenchymal stem cells and define appropriate routes for their therapeutic implementation.

Candidates must hold a PhD in a relevant subject and be highly motivated scientists with interests in stem cell biology and routes to controlling their differentiation and therapeutic applications. The project uses a variety of cell biology, molecular biology and biochemical techniques. Experience in ES cell culture is desirable, but training can be provided.

Salary will be £25,134 per annum. This post is available immediately and will be offered on a fixed-term contract for a period of one year.
